The display has no scaler, so it'll just take the pixel data given and display it. That is in common with almost any DPI, DSI, or SPI display, and a moderate number of cheaper HDMI displays.
The display is 720x1280, and the pixel data given to it needs to be 720x1280. (Same was true of the old display that it was 800x480 and had to be given 800x480).

Adding video=...,rotate=270 only rotates the rendering of the console text, not the framebuffer itself (the Pi SoC doesn't support realtime transpose).
