Yes you can...however you will need an adaptor. Be aware that there are issues with some adaptors on the PI4...There's a sticky that talks about it...
Bigger SD cards might be a cheaper option too....On our "desktop" 400s we use Samsung evo 64GB SD cards as our "working" drives. They work great in that role. I also have SSDs and USB sticks and also large SR drives, which are used for various roles. They all work OK and all have their pros and cons...It depends what you want to do.
I would always advise storing data separately from your boot drive if you can though...and of course; doing regular backups...and if your data is important to you; offsite storage of some sort.
